Effect of Acute Ipobaric Hypox a on the Defense Mechanisms Against Oxygen Toxicity in Rat Heart

Abstract
The possibility that acute hypobaric-hypoxia alters the defense mechanisms against oxygen toxicity in rat heart has been investigated. Four different degrees of acute hypobaric-hypoxia were induced in vivo by hypobaric chamber flight for 2 hours at 15OOOft, 20,000ft,25,000ft and 30,000ft. At the end of the each altitude exposure,the changes in serum ATP ,lactate and glucose were determined. And the productions of superoxide radical and malondialdehyde(MDA),and the activities of total superoxide dismutase (total SOD),glutathione peroxidase (GSH-Px)and total ATPase in heart homogenates have been determined. Both serum and tissue ATP and glucose were decreased according to altitude. And serum and tissue lactate were increased according to altitude. Acute hypobaric-hypoxia enhanced the productions of superoxide radical and MDA in heart homogenate as much as 125.8 % and 246.9% than normal value, respectively, when altitudes ranged from 15,000 to 30,000ft. Also the activities of total SOD,GSH-Px and total ATPase. in heart homogenate showed 29.7%,22.1% and 21.5 % inhibitions compared with normal value at 30,000ft, respectively These results suggest that high levels of superoxide radical and pe.roxidized lipid resulting from inhibited superoxide dismutase and glutathione peroxidase may play a role in acute hypobaric-hypoxia induced heart damage.
